KEY LEARNING OBJECTIVES
Post translational modifications (PTMs) of protein biopharmaceuticals, such as, glycosylation, oxidation, phosphorylation, sulfation, lipidation, disulfide bond formation, and deamidation, can affect their function and stability. In most cases these chemical changes occur in vivo but some can also occur in vitro: for example, during various stages of manufacture such as purification and storage. Assessing PTMs is complex, as many biopharmaceuticals bear a combination of two or more PTMs. This webcast will examine recent advancements in chromatography and mass spectrometry for full and accurate post translation modification characterization of protein biopharmaceuticals. This will include examining the use of reversed phase HPLC and mass spectrometry to detect and investigate protein modifications by monitoring mass differences and analyzing peptide fragments and their mass alterations. The impact of analysis method on PTMs will also be discussed with alternatives to classical collisional-induced dissociation MS fragmentation methods being considered (electron transfer dissociation (ETD) and electron capture dissociation (ECD)). We will also consider the application of LC-MS for host cell protein quantitation and determination of antibody-drug conjugates in biological matrices.
